Recombination events are not uniformly distributed and often cluster in narrow regions known as recombination hotspots. Several studies using different approaches have dramatically advanced our understanding of recombination hotspot regulation. Population genetic data have been used to map and quantify hotspots in the human genome. Genetic variation in recombination rates and hotspots usage have been explored in human pedigrees, mouse intercrosses, and by sperm typing. These studies pointed to the central role of the PRDM9 gene in hotspot modulation. In this study, we used single nucleotide polymorphisms (SNPs) from whole-genome resequencing and genotyping studies of mouse inbred strains to estimate recombination rates across the mouse genome and identified 47,068 historical hotspots--an average of over 2477 per chromosome. We show by simulation that inbred mouse strains can be used to identify positions of historical hotspots. Recombination hotspots were found to be enriched for the predicted binding sequences for different alleles of the PRDM9 protein. Recombination rates were on average lower near transcription start sites (TSS). Comparing the inferred historical recombination hotspots with the recent genome-wide mapping of double-strand breaks (DSBs) in mouse sperm revealed a significant overlap, especially toward the telomeres. Our results suggest that inbred strains can be used to characterize and study the dynamics of historical recombination hotspots. They also strengthen previous findings on mouse recombination hotspots, and specifically the impact of sequence variants in Prdm9.

Abstract This study uses global positioning systems (GPS) technology to compare and contrast the behaviour patterns of first-time and repeat visitors to Hong Kong. The use of GPS recorders, coupled with an analysis using Geographical Information System (GIS) software produces highly accurate, fine-grained information concerning the spatial and time?space patterns of the visitors. The study findings build on existing knowledge that first-timers tend to travel more widely throughout the destination while repeat visitors tend to confine their actions to a smaller number of locations. However, the data also revealed that first-timers and repeaters spend different amounts of time at the same attractions and visit during different times of the day. First-time visitors also tend to make one long, extended day trip from the hotel, while repeaters tend to make a number of shorter forays, returning to the hotel intermittently during the day. The social relevance of framing effects hinges upon their ability to persist. This article develops a theoretical account of the conditions under which framing effects should vanish quickly, fade slowly, or cause permanent changes. It argues that the cognitive processes involved in mediating frame effect need to leave durable traces in a person’s knowledge to raise a persistent effect. This paper distinguishes temporary changes in the accessibility of knowledge from durable changes in the applicability structure and belief content. It discusses under which conditions these memory traces will likely affect judgment formation also after the stimulus is gone. We argue that the durability of framing effects can be modeled based on the chronic accessibility of frame-relevant knowledge and the familiarity of the frame.

This paper suggests a model of obtaining estimates of capital stock based on the theory of ‘flexible accelerator’. However, this represents a rather ‘indirect’ method independently for each year and each region. Clearly this is an unrealistic condition, especially for regional economies characterized by mutual spatial dependence. To add an extra injection of realism, we illustrate how a national model of capital stock (the stock –flow model) can effectively be ‘regionalized’

Flash floods caused by convective rain storms are highly sensitive to the space–time character- istics of rain cells. In this study we exploit the high space–time resolution of the radar data to study the characteristics of the rain cells and their impact on flash flood magnitudes. A rain cell model is applied to the radar data of an actual storm and the rain fields represented by the model further serve as input into a hydrological model. Global sensitivity analysis is applied to identify the most important factors affecting the flash flood peak discharge. As a case study we tested an extreme storm event over a semi-arid catchment in southern Israel. The rain cell model was found to simulate the rain storm adequately. We found that relatively small changes in the rain cell’s location, speed and direction could cause a three-fold increase in flash flood peak discharge at the catchment outlet.

We experimentally demonstrate a frequency modulation locked servo loop, locked to a resonance line of an on-chip microdisk resonator in a silicon nitride platform. By using this approach, we demonstrate real-time monitoring of refractive index variations with a precision approaching 10−7  RIU, using a moderate Q factor of 104. The approach can be applied for intensity independent, dynamic and precise index of refraction monitoring for biosensing applications.

Transgenic mice expressing the diphtheria toxin receptor (DTR) in specific cell types are key tools for functional studies in several biological systems. B6.FVB-Tg(Itgax-DTR/EGFP)57Lan/J (CD11c.DTR) and B6.Cg-Tg(Itgax-DTR/OVA/EGFP)1Gjh/Crl (CD11c.DOG) mice express the DTR in CD11c(+) cells, allowing conditional depletion of dendritic cells. We report that dendritic-cell depletion in these models caused polymorphonuclear neutrophil (PMN) release from the bone marrow, which caused chemokine-dependent neutrophilia after 6-24 h and increased bacterial clearance in a mouse pyelonephritis model. We present a transgenic mouse line, B6.Cg-Tg(Itgax-EGFP-CRE-DTR-LUC)2Gjh/Crl (CD11c.LuciDTR), which is unaffected by early neutrophilia. However, CD11c.LuciDTR and CD11c.DTR mice showed late neutrophilia 72 h after dendritic cell depletion, which was independent of PMN release and possibly resulted from increased granulopoiesis. Thus, the time point of dendritic cell depletion and the choice of DTR transgenic mouse line must be considered in experimental settings where neutrophils may be involved.
