Meat products contain valuable nutrients that are important for human health and development but are also highly susceptible to colonization by microorganisms. This can lead to spoilage and serious foodborne illnesses. Natural antimicrobial peptides, produced by many organisms as part of their innate immune system to fight microbial infections, have great potential as food preservatives. In this study, we explored the effect of ternary antimicrobial random peptide mixtures (RPMs) on food spoilage bacteria in minced turkey meat. Amendment of RPMs to meat led to significant reductions in bacterial abundance in experimental tests, and RPMs worked synergistically with nitrite to reduce bacterial loads. Using high-throughput 16S ribosomal RNA gene amplicon sequencing, we characterized the effect of RPMs and nitrite on meat microbial community structure before and during incubation under refrigerated conditions. Our findings reveal strong antimicrobial activity for RPMs against spoilage bacteria in meat, including Listeria monocytogenes and Pseudomonas putida. These results demonstrate the potential of RPMs as a safer preservative for reducing spoilage in meat and other food products.

As the male reproductive organ, the main task of the testis is the production of fertile, haploid spermatozoa. This process, named spermatogenesis, starts with spermatogonial stem cells, which undergo a species-specific number of mitotic divisions until starting meiosis and further morphological maturation. The pituitary gonadotropins, luteinizing hormone, and follicle stimulating hormone, are indispensable for vertebrate spermatogenesis, but we are still far from fully understanding the complex regulatory networks involved in this process. Therefore, we developed an testis cultivation system which allows evaluating the occurring changes in histology and gene expression. The experimental circulatory flow-through setup described in this work provides the possibility to study the function of the male tilapia gonads on a cellular and transcriptional level for at least 7 days. After 1 week of culture, tilapia testis slices kept their structure and all stages of spermatogenesis could be detected histologically. Without pituitary extract (tilPE) however, fibrotic structures appeared, whereas addition of tilPE preserved spermatogenic cysts and somatic interstitium completely. We could show that tilPE has a stimulatory effect on spermatogonia proliferation in our culture system. In the presence of tilPE or hCG, the gene expression of steroidogenesis related genes ( and ) were notably increased. Other testicular genes like , or were not expressed differentially in the presence or absence of gonadotropins or gonadotropin containing tilPE. We established a suitable system for studying tilapia spermatogenesis with promise for future applications.

Oxidative stress (OS) and excessive reactive oxygen species (ROS) production have been implicated in many neurological pathologies, including acute seizures and epilepsy. Seizure-induced damage has been demonstrated both in vitro and in several in vivo seizure and epilepsy models by direct determination of ROS, and by measuring indirect markers of OS. In this manuscript, we review the current reliable methods for quantifying ROS-related and OS-related markers in pre-clinical and clinical epilepsy studies. We first provide pieces of evidence for the involvement of different sources of ROS in epilepsy. We then discuss general methods and assays used for the ROS measurements, mainly superoxide anion, hydrogen peroxide, peroxynitrite, and hydroxyl radical in in vitro and in vivo studies. In addition, we discuss the role of these ROS and markers of oxidative injury in acute seizures and epilepsy pre-clinical studies. The indirect detection of secondary products of ROS such as measurements of DNA damage, lipid peroxidation, and protein oxidation will also be discussed. This review also discusses reliable methods for the assessment of ROS, OS markers, and their by-products in epilepsy clinical studies.

This chapter considers two cases of contemporary genizah in terms of analog and digital media as they comply and conflict with traditional decrees. The first is a debate surrounding the discarding of audiocassettes containing sermons that if were to be written textually rather than sound recorded would have to be treated as genizah worthy. The second is the feasibility of an obligatory digital genizah, containing both digital artifacts and documents. Ultimately, both cases were not deemed as genizah worthy due to their non-textual and non-inscriptional logic.

Few studies have focused on the Aramaic of the Zohar, and to this day, only one of these presents a completed grammatical analysis. Scholars have dealt at large, however, with the question of whether the Aramaic of the Zohar is artificial or not. I briefly review the history of the literature around this question, then propose my own criteria to examine whether a language of a given text is indeed artificial. Finally, I put this methodology into practice, as I investigate the nature of Zoharic Aramaic by examining specific linguistic phenomena in the relevant corpus.

Costly investor-state dispute settlement (ISDS) cases have led several developing countries to take far-reaching steps to distance themselves from the global investment regime, such as the denunciation or renegotiation of international investment agreements (IIAs) or the withdrawal from the International Centre for Settlement of Investment Disputes (ICSID). Despite facing the highest number of investment claims worldwide and despite being very vocal about the shortcomings of the current regime, Argentina has neither denounced a single IIA nor renounced ICSID. This article addresses this puzzle. It first shows that under the Kirchners’ governments (2003-2015), Argentina adopted a dual approach: maintaining its IIAs and membership in ICSID on the one hand, but vigorously fighting ISDS awards on the other. Using in-depth interviews, news reports, and secondary sources, it then demonstrates that this ‘neither-in-nor-out’ approach is best explained by a unique Argentinian identity, which combines Latin American and Western dimensions, conditioned by external political and economic constraints, especially in the immediate aftermath of the 2001 financial crisis. As such, this study underscores the need to account for both material and ideational factors when striving to grasp development-related foreign policy.

Schizophrenia is a mental disease that results in decreased life expectancy and well-being by promoting obesity and sedentary lifestyles. Schizophrenia is treated by antipsychotic drugs. Although the second-generation antipsychotics (SGA), Olanzapine and Aripiprazole, are more effective in treating schizophrenia, they display a higher risk of metabolic side effects, mostly by development of diabetes and insulin resistance, weight gain, and dyslipidemia. Endoplasmic reticulum (ER) stress is induced when ER homeostasis of lipid biosynthesis and protein folding is impaired. This leads to the activation of the unfolded protein response (UPR), a signaling cascade that aims to restore ER homeostasis or initiate cell death. Chronic conditions of ER stress in the liver are associated with diabetes and perturbed lipid metabolism. These metabolic dysfunctions resemble the pharmacological side effects of SGAs. We therefore investigated whether SGAs promote the UPR in human and mouse hepatocytes. We observed full-fledged activation of ER stress by Aripiprazole not by Olanzapine. This occurred at low micromolar concentrations and to variable intensities in different cell types, such as hepatocellular carcinoma, melanoma, and glioblastoma. Mechanistically, Aripiprazole caused depletion of ER calcium, leading to activation of inositol-requiring enzyme 1 (IRE1)and protein kinase R (PKR)-like endoplasmic reticulum kinase (PERK), two major transducers of the UPR. Cells underwent apoptosis with Aripiprazole treatment, which coincided with UPR induction, and this effect was reduced by adding glutathione without affecting UPR itself. Deletion of IRE1 from HepG2, a human liver cancer cell line, protected cells from Aripiprazole toxicity. Our study reveals for the first time a cytotoxic effect of Aripiprazole that involves the induction of ER stress. SIGNIFICANCE STATEMENT: The antischizophrenic drug Aripiprazole exerts cytotoxic properties at high concentrations. This study shows that this cytotoxicity is associated with the induction of endoplasmic reticulum (ER) stress and IRE1 activation, mechanisms involved in diet-induced obesity. Aripiprazole induced ER stress and calcium mobilization from the ER in human and mouse hepatocytes. Our study highlights a new mechanism of Aripiprazole that is not related to its effect on dopamine signaling.

Located in the theoretical triangle of urban, cultural, and political studies, this paper examines artists’ political engagement in cities. Applying a qualitative approach through inter-urban fieldwork in Hamburg and Hanover, Germany, and Jerusalem and Tel Aviv-Jaffa, Israel, we study how artists critically engage with their cities and urban spaces, and analyze how the artists’ themselves perceive and practice their own actions. Based on this, we offer a typology of artists’ attitudes and behavior patterns toward urban politics and policies, which we propose as a relevant theoretical tool for analyzing political engagement of artists in the urban environment.
